10X TB Salts 1000 ml (ONE BOTTLE TO BE MADE FOR EVERY 10 BOTTLES OF TB)

• Start with 600 ml of ddH₂O

• Add 23.1 g KH₂PO₄ (potassium phosphate monobasic)

• Add 125.4 g K₂HPO₄ (potassium phosphate dibasic)

• Add ddH₂O to 1000 ml

• Label bottle "10X TB SALTS" with your initials and the date

• Autoclave

• Start with 600 ml of ddH₂O
• Add 12 g Bacto Tryptone

• Add 24 g Yeast Extract

• Add 4 ml glycerol

• Add ddH₂O to 900 ml (NOT 1000 ml!)

• Label bottles "TB" with your initials and the date

• Autoclave, cool

• Working in the hood, add 100 ml of 10X TB Salts to a bottle of cooled TB (the volume may be less than 900 ml due to loss during autoclaving)
• Adjust the final volume to 1 liter with sterile ddH₂O
• Leave the original label on the bottle and add a new label beneath it
• Label bottle "TB+SALTS" with your initials and the date

• Add 1.5 ml Ampicillin stock (100 mg/ml) to 1000 ml TB/S

• Start with 800 ml ddH₂0
• Add 10 g Bacto Tryptone
• Add 5 g Yeast Extract
• Add 10 g NaCl
• Adjust pH to 7.5
• Add ddH₂0 to 1000 ml
• Do not put more than 900 ml into 1000 ml bottle
• Label bottles "LB BROTH" with your initials and date
• Autoclave

• Follow exact recipe for LB
• Add 7.5 g agar to each 500 ml in a 1000 ml bottle
• Label bottles "LB-AGAR" with your initials and the date
• Autoclave

• Start with 800 ml ddH₂0
• Add 10 g NZ Amine
• Add 5 g NaCl
• Add 5 g Yeast Extract
• Add 2 g MgSO_{4 ·} 7H₂0
• Adjust pH to 7.0
• Add ddH₂0 to 1000 ml
• Do not put more than 900 ml into 1000 ml bottle
• Label bottles "NZY BROTH" with your initials and the date
• Autoclave

• Follow exact recipe for NZY
• Add 7.5 g agar to each 500 ml in a 1000 ml bottle
• Label bottles "NZY-AGAR" with your initials and the date
• Autoclave

• Follow exact recipe for NZY
• Add 3.5 g of agarose to each 500 ml in a 1000 ml bottle
• Label bottles "NZY TOP AGAROSE" with your initials and the date
• Autoclave

• Start with 50 ml of sterile ddH₂O
• Add 10 g Maltose
• Add sterile ddH₂O to 100 ml
• Slightly warm solution to fully dissolve Maltose
• Filter sterilize

• Start with 450 ml of sterile ddH₂O
• Add 12.3 g MgSO_{4 ·} 7H₂O
• Add sterile ddH₂O to 500 ml
• Slightly warm solution to fully dissolve MgSO₄
• Filter sterilize
