Freezing Medium

To prepare ~200 ml (all components should already be sterile, and the mixture should be prepared in a sterile hood):

Start with 180 ml TB broth
Add 20 ml 10× salts
Add 14 ml of previously autoclaved 100% glycerol (use a syringe)
Add 321 μl of 100 mg/ml ampicillin (do not add ampicillin to hot medium and never add it before autoclaving)

Alternatively, if TB + salts is already available, 200 ml of TB + salts can be substituted for the BT broth and 10× salts in the above recipe.
