Sorghum EST Libraries

An EST (expressed sequence tag) database has been assembled from a variety of sorghum cDNA libraries. These libraries were prepared from diverse tissues and developmental stages, and in response to a variety of external stimuli such as drought or hormone treatment. For most libraries, approximately 5000 cDNAs were sequenced from both 3' and 5' ends.

All libraries are non-amplified. Bacterial clones corresponding to all sequenced cDNAs have been archived and are available upon request, as are any remaining aliquots of the various libraries.

A non-redundant cDNA collection (Milestone Unigene Set) has been assembled and currently numbers close to 16,000 members. It is being used for expression profiling utilizing microarrays on glass microslides.

The first 10 libraries were prepared from the mapping parent BTx623. For these libraries, cDNA prepared from polyA+ RNA was cloned into lambda ZAP II. pBluescript SK(-) was obtained by mass excision.

Subsequent libraries are being prepared through a collaboration with Drs. Sumio Sugano and Yutaka Suzuki of the Tokyo University College of Medicine from RNA prepared by Robert Klein, one of the co-PIs on our NSF Plant Genome project. They have developed excellent technology for preparing libraries highly enriched in full-length cDNAs. These libraries have been prepared from a variety of genotypes to facilitate detection of genetic polymorphisms.
